Description
Starting in 2002, Jens Peter Fürste et al. screened high-affinity RNA aptamers against betaA4(1-40), providing a potential tool for applications in the analysis and diagnosis of Alzheimer's disease. Subsequently, in 2009, Hisakazu Mihara et al. screened RNA aptamers against Abeta1-40 using the systematic evolutionary ligand's exponential enrichment (SELEX) procedure, which were effective in inhibiting the aggregation of Abeta and preventing its fibrillization, demonstrating the potential to be used as therapeutic agents for Alzheimer's disease. In the same year, Gal Bitan et al. screened aptamers that, although unable to specifically recognize Abeta40 oligomers, were responsive to a wide range of amyloid fibrils and showed a high degree of sensitivity for the detection of amyloid β-fold structures, suggesting that they have the potential to be used as a highly efficient amyloid recognition tool. This series of research advances not only deepens our understanding of the application of aptamers in amyloid research, but also provides new ideas and approaches for the diagnosis and treatment of Alzheimer's disease[1].SELEX
Jens Peter Fu¨rste et al. isolated a high-affinity RNA aptamer for β-amyloid peptide (βA4(1-40)) targeting Alzheimer's disease from a combinatorial library containing about 10^15 different molecules by an in vitro screening technique (SELEX). A DNA library containing 70 random nucleotide sequences was used, amplified by PCR and transcribed into an RNA library. β amyloid peptide was covalently coupled at the N-terminus via an additional cysteine residue to thiopropyl-activated Sepharose 6B, and the specifically bound RNA was eluted by DTT, reverse transcribed into cDNA and amplified by PCR for eight rounds of screening. The screened RNA aptamers were cloned, amplified, and sequenced, and four major conserved secondary structure modalities were found, of which the β55 aptamer had the lowest dissociation constant (Kd) of 29 nM. The dissociation constants of the aptamer with β-amyloid peptide were determined by analytical affinity chromatography and ranged from 29-48 nM[1].

Structure
The sequence and secondary prediction structure of the aptamer will be shown here, here we used ribodraw to complete the figure. In addition,The authors also used the dissociation constants of the aptamer with β-amyloid peptide were determined by analytical affinity chromatography and ranged from 29-48 nM. By this method, the researchers were able to accurately determine the dissociation constant (Kd) between the two, which was determined to be 29-48 nM. The 2D structure of the figures is based on the prediction results of the RNA fold website by ribodraw tool to draw.
